Documented hypersensitivity (allergy) to dolutegravir, emtricitabine, tenofovir alafenamide, or any of the tablet’s excipients. Severe renal impairment (creatinine clearance < 30 mL/min) — as TAF’s safety may be compromised. Severe hepatic impairment (Child-Pugh C) — use not recommended. Also, caution/avoid combining with certain drugs (for e.g. dofetilide, due to risk of harmful interactions) as per prescribing information.
Side Effects:
Common side effects may include: HeadacheNauseaDiarrhea / gastrointestinal upsetFatigue, tirednessInsomnia or sleep disturbancesPossible serious but less common risks / concerns: Renal effects — Impaired kidney function, particularly in those with pre-existing renal disease. Liver toxicity / worsening hepatic disease (especially if concurrent hepatitis or liver problem) Bone mineral density reduction / risk of bone-related issues (some risk with tenofovir analogues) Immune reconstitution inflammatory syndrome (IRIS) — in some patients, as immunity recovers, latent infections may flare. Hypersensitivity reactions (rare but possible) — allergic reactions, organ dysfunction
Precautions:
Must be taken once daily at same time — consistency is key. Missing doses can reduce effectiveness or lead to resistance. Use with caution in patients with renal or hepatic impairment — discuss with doctor. In presence of co-infections (e.g. hepatitis B or C), close monitoring is needed — drug may impact liver, and HBV reactivation possible if therapy changed. Bone health — long-term use may affect bone mineral density — especially relevant in adolescents, older adults or people with risk factors. Adherence matters — irregular intake, stopping/restarting without guidance can cause viral rebound, resistance, loss of efficacy. Pregnancy / breastfeeding: Use should be under specialist supervision; there may be special considerations.
Dietary Restrictions:
Dobataf-3 can be taken with or without food. Maintain good hydration, especially if side effects like nausea or diarrhea occur.Avoid substance abuse (alcohol, recreational drugs) — can stress liver or kidneys, interfere with immune function and adherence.Maintain balanced diet, adequate nutrition — important for immune system and general health along with ART.Bone health support — ensure adequate calcium and vitamin D (per doctor’s advice), especially if treatment is long-term or bone risk factors exist.
